INQUIRYIntroductions
Dot blot hybridization uses the principle of nucleic acid hybridization to detect whether there is nucleic acid hybridization and its hybridization strength. It is mainly used for the detection of plant gene deletion or copy number variation. Its experimental principle is very simple. Firstly, the DNA to be tested is denatured and spotted on a nitrocellulose membrane (or nylon membrane), bake and fix it; Secondly, hybridization is performed with a labeled probe; Then the membrane is washed to remove unattached probes; Finally, autoradiography. This technology is mainly used for the detection of gene deletion or copy number change.
