Chestnut ink disease is a devastating disease of chestnut worldwide caused by the Phytophthora. Root rot, caused by Phytophthora cambivora, primarily affects the trunk and roots, resulting in decay and black discharge. The disease is now widely distributed in chestnut-growing areas in Europe and North America, and is particularly damaging in old chestnut forests at high altitudes. Symptoms of Chestnut Ink Disease

  • Root lesions. Initial necrosis of the primary or lateral roots with flaming brown to black lesions in the cortex, accompanied by black exudate.
  • Stem base symptoms. Epidermal ulceration in the area below 1 meter of the trunk, thickening of the lenticels, and discharge of dirty black sap, later root rot, and death of the whole plant.
  • Above-ground part of the performance. Due to root damage, leaf yellowing, wilting, branch cracking, tree weakness, and in severe cases, the whole plant dies.

Genetic transformation of chestnut

We utilize somatic cell embryos as receptors in combination with Agrobacterium-mediated gene transfer methods.

The pair includes optimizing a system for efficient regeneration of chestnut somatic cell embryos, such as screening key transcription factors to regulate the developmental process of somatic cell embryos. The establishment of a hairy root transformation system in combination with Agrobacterium rhizogenes-mediated transformation methods will also provide a tool for the functional validation of root disease resistance genes. In addition, we established a highly efficient transformation mediated by Agrobacterium tumefaciens in chestnut. We achieved stable expression of target genes in chestnut tissues by optimizing somatic embryo induction, co-culture conditions, and screening markers, with a substantial increase in transformation efficiency compared with traditional methods. Our chestnut genetic transformation technology lays the foundation for screening ink disease resistance genes.

Gene editing technology

We have established a gene editing technology platform to precisely edit resistance-related genes to enhance defense against Phytophthora, which can be used for chestnut ink blight control research. In addition, we can edit multiple resistance loci simultaneously by using multiple sgRNA designs, such as integrating anti-fungal and anti-insect genes, in response to the composite threat of ink disease with root rotting molds and insect pests.

Transgenic technology

Transgenic technology in chestnut disease resistance has evolved from transferring a single resistance gene to integrating multiple strategies. For example, the cross-species transfer of exogenous disease resistance genes into American chestnut has successfully conferred resistance to chestnut blight, and similar strategies can be extended to chestnut ink disease control. In addition, natural resistance genes within the chestnut genus are preferentially selected to mimic the effects of traditional hybridization through overexpression or tissue-specific promoter drive. For example, a 35S strong promoter was used to drive high expression of disease resistance genes in the root system, targeting defense against root infestation by ink blight.
