Medicago truncatula Transformation
INQUIRYIntroductions
Barrelmedic is an annual plant in the genus Legume and Medicago sativa. The Latin name is Medicago truncatula.
Its ploidy is small (2n=16), and its genome is also small (450-500 Mbp).
It is self-pollinated and has more seeds.
Its growth cycle is short (about 3 months).
There are a large number of mutants and multiple ecotypes.
It has high biodiversity.
Its genetic transformation efficiency is high.
It has high genetic similarity with most legumes.
Because Medicago truncatula has these advantages, it is considered as a model plant for studying legume genetics. It can not only be used in the study of legume biology, but also promote other factors such as human nutrition, plant pathogenic response, root development, symbiotic interaction, metabolism of carbon, nitrogen and phosphorus in plants and the hormone control and signal transduction of plant development.
Services
Lifeasible has established a Medicago truncatula transformation platform through Agrobacterium-mediated method method. The platform has a complete, large-scale, standardized technical system, and provides professional and convenient technologies for barrelmedic genetic transformation for agricultural personnel who serve barrelmedic as the research object. We can provide customers with barrelmedic genetic transformation services, including experimental design, vector construction, plasmid transformation, positive transplantation screening and testing, etc. We have an experienced team that can help you solve technical problems and challenges in barrelmedic conversion to help your project succeed.
Customer provided materials
- The sequence of the target gene and the Gene Bank ID
- Plasmids, Agrobacterium, etc.
- Vector to be transformed (conventional vector can also be provided by us)
- Unconventional receptor material
※ We do not accept the transformation service of lethal genes or severely defective genes. If there are any of the above, please inform our company in advance.
Receptor material
- Recipient material varieties: R108-1 and Jemalong A17.
- Recipient material types: leaf explants, root explants.
Transformation methods
- Using leaf or root explants derived from the R108-1 genotype with strong regenerative ability.
- Using cotyledon explants from Jemalong A17 genotype, allowing rapid propagation through direct shoot organogenesis.
- Via vacuum infiltration flowering plants or seedlings developed from the Arabidopsis "floral dip" method, which requires no plant tissue culture process.

Feedback to customers
- Complete experimental methods, original experimental records, instrument parameters, reagents and consumables information
- Some pictures of genetic transformation and screening
- Picture of some samples of callus differentiation and regeneration
- Agarose gel electrophoresis image of foreign gene PCR detection of T0 generation transgenic seedlings
- T0 generation transgenic vaccine: If we use the vector and recipient provided by us, the T0 generation positive vaccine for each transformation experiment is ≥12; if the customer needs to use other vectors and recipients, we usually only promise to get the T0 generation positive vaccine 6-8 strains.
Service process
Project cycle
It takes about 3-4 months from the construction of the vector to the test results of the T0 generation transgenic seedlings. And experiments can be carried out throughout the year.
Advantages
- When using cotyledon or root explants, shoot regeneration can be carried out directly without the process of callus induction, so it is more convenient and quicker.
- The method of transfecting alfalfa inflorescence by the method similar to that of Arabidopsis does not require tissue culture process, so the experiment period can be shortened.
- Many different antibiotic markers (such as Kanamycin, Hygromycin and Phosphinothricin) are available for your choice.
- A variety of Agrobacterium strains are available for you to choose, such as GV3101, AGL-1, EHA105 and C58C1.
- Rich project execution experience and leading research strength.
- Perfect service support. Provide professional technical consultation and guidance in the early stage to optimize the project; communicate at any time during the experiment to keep abreast of the progress; after the project is completed, provide complete results according to the contract.
