INQUIRYIntroductions
Biochemical markers mainly use gene expression products such as isoenzymes and storage proteins to reflect genotype differences to a certain extent. They are used in the genetic breeding of wheat, corn and other crops. Isozyme refers to a group of enzymes that catalyze the same chemical reaction, but have obvious differences in protein molecular structure, physical and chemical properties, and immune performance. This enzyme exists in the same genus of organisms, or in different tissues/cells of the same individual. The production of isoenzymes is due to the adaptation to metabolic processes during biological evolution. For enzymes that are more critical for metabolic regulation or closely related to the external environment, they also have isoenzyme properties and are more common, and at the same time there are more molecular types. Through the analysis of isoenzyme profiles, we can study a variety of information such as species evolution, genetic variation, hybrid breeding, and individual development differences. Therefore, this research is widely used in the field of plant molecular breeding.

Technical process
- • First, the samples were separated by SDS-polyacrylamide electrophoresis (SDS-PAGE, 0.1% gelatin).
- • Then the enzyme in the sample is restored to activity in a buffer system containing divalent metal ions.
• The gelatin in the gel is hydrolyzed at the respective migration positions. -
• Finally, dye the gel with the corresponding dye and decolorize the gel.
• The strength of the band is proportional to the enzyme activity.
Analysis content
We use electrophoresis analysis method to analyze the isozyme spectrum, and draw the isozyme spectrum based on the difference of electrophoretic mobility.
