Brassica napus Transformation
INQUIRYIntroductions
Rapeseed, whose scientific name is Brassica napus L., is a herbaceous crop of the cruciferous family and Brassica genus. It is native to Europe and is widely grown all over the world. After multiple generations of propagation and hybridization, many varieties with different characteristics and adapted to different growth conditions have been cultivated, mainly including Brassica napus, Brassica Juncea and Brassica rape. Rapeseed is one of the four major oil crops and ranks among the five major economic crops in the world. It is an important source of edible vegetable oil and protein. With the rapid development of biotechnology, especially the emergence of gene recombination and transgenic technology, a new path has been opened up for the genetic improvement of rape.
Services
Lifeasible has established a rape genetic transformation platform, using Agrobacterium-mediated method or gene gun method to provide large-scale and standardized transformation technology to realize rape genetic transformation. Customers can provide target genes, plasmids, Agrobacterium, etc., and we will complete the entire experimental process from constructing vectors to obtaining transgenic vaccines.
Customer provided materials
- If only gene sequence information (or Gene Bank ID) is provided, our company can undertake gene amplification and vector construction services, which are charged separately.
- Constructed Escherichia coli vector plasmid, but provide clear resistance information for screening.
- Unconventional receptor material
※ We do not accept the transformation service of lethal genes or severely defective genes. If there are any of the above, please inform our company in advance.
Brassica napus transformation methods
- Agrobacterium-mediated transformation
- Gene gun bombardment method
Receptor material
- Recipient species
- Recipient types
Westar, Qingza5 (305), Xiangyou15, Zhongshuang11 and other varieties
Cotyledon, hypocotyl or seedling
Workflow of rape transformation by Agrobacterium-mediated method
- Vector construction
- Rape receptor material cultivation
- Agrobacterium-mediated genetic transformation and screening
- Differentiation of positive callus into seedlings
- Strong plantlets and rootage of regenerated green seedlings
- PCR detection of foreign genes in T0 generation transgenic seedlings
Feedback to customers
- Complete experimental report, including methods, original experimental records, instrument parameters, reagents and consumables information.
- Some pictures of genetic transformation and screening.
- Picture of some samples of callus differentiation and regeneration.
- Agarose gel electrophoresis image of foreign gene PCR detection of T0 generation transgenic seedlings.
- T0 generation transgenic vaccine: If we use the vector and recipient provided by us, there will be 8-10 T0 generation positive vaccines for each transformation experiment; if customers need to use other vectors and recipients, we usually only promise to get T0 generation positive vaccines 6-8 strains.
Service process
Project cycle
- It takes about 4~6 months.
Advantages
- Agrobacterium-mediated transformation is fast, efficient, and stable, and mainly single-copy insertion.
- For commonly used rape varieties, sufficient sterile seedlings are prepared all year round, and transformation experiments can be carried out at any time to save customers' time.
- We have a variety of labeled vectors and media to provide you with a wide range of choices.
- The technical team engaged in plant genetic transformation for many years guarantees the success rate of the experiment.
- Personalized customized package service.
