MeRIP-seq in Plant
INQUIRYIntroductions
It is known that there are more than 100 modifications in RNA, and m6A methylation is the most common modification in eukaryotic mRNA. m6A is mainly enriched in the promoter region and stop codon region of mRNA. MeRIP-seq (Methylated RNA Immunoprecipitation Sequencing) is based on the principle that m6A-specific antibodies recognize and bind to m6A on mRNA. It is based on the enrichment of methylated modified fragments by RNA immunoprecipitation method and combined with high-throughput sequencing to study the region of mRNA where m6A methylated modification occurs. MeRIP-seq has been widely used in the field of agricultural breeding, mainly for the study of eukaryotic RNA methylation distribution, the study of plant mRNA methylation and the response mechanism of environmental stress, the study of mRNA anti-degradation mechanism, etc.

Sample requirements
- Cell samples or RNA samples of plant tissues
1) If the customer conducts the IP experiment by himself, the single library construction requires RNA after IP is: RNA ≥ 20ng, concentration ≥ 10 ng/µl, fragment < 300nt. And it does not contain EDTA, no DNA contamination (it is recommended to meet the amount of at least 2 library building).
2) If you need to submit the IP experiment to us, the sample size required for a single IP experiment is: RNA ≥ 300μg (plant tissue sample ≥ 2g), concentration > 350ng/µl, no EDTA, no DNA contamination. It is recommended to send 2~3 times the sample size for backup.
