Introductions

MeRIP seq in Plant

It is known that there are more than 100 modifications in RNA, and m6A methylation is the most common modification in eukaryotic mRNA. m6A is mainly enriched in the promoter region and stop codon region of mRNA. MeRIP-seq (Methylated RNA Immunoprecipitation Sequencing) is based on the principle that m6A-specific antibodies recognize and bind to m6A on mRNA. It is based on the enrichment of methylated modified fragments by RNA immunoprecipitation method and combined with high-throughput sequencing to study the region of mRNA where m6A methylated modification occurs. MeRIP-seq has been widely used in the field of agricultural breeding, mainly for the study of eukaryotic RNA methylation distribution, the study of plant mRNA methylation and the response mechanism of environmental stress, the study of mRNA anti-degradation mechanism, etc.

Services

With years of rich experience, Lifeasible provides methylated RNA immunoprecipitation sequencing (MeRIP-seq) and analysis services. Customers only need to provide tissues or RNA sample, and we complete the entire service process from sample extraction, quality testing, sequencing, biological information analysis to making reports.

Analysis content

Analysis content Solved problem
Quality assessment of sequencing data Filter out low-quality data to ensure data quality
Alignment analysis of reference sequence Statistics of reads distribution on the only aligment
Peak calling Detect the credible area where m6A modification occurs
Motif analysis Preference of protein binding sequence
Related gene annotation of peak Peak in the genome and transcripts
Difference peak analysis Analyze the difference between different samples peak
Related gene function analysis GO/KEGG enrichment analysis of peak related genes
Personalized analysis, multi-omics association analysis (need to contact us for evaluation)

Sample requirements

  • Cell samples or RNA samples of plant tissues
  • 1) If the customer conducts the IP experiment by himself, the single library construction requires RNA after IP is: RNA ≥ 20ng, concentration ≥ 10 ng/µl, fragment < 300nt. And it does not contain EDTA, no DNA contamination (it is recommended to meet the amount of at least 2 library building).

    2) If you need to submit the IP experiment to us, the sample size required for a single IP experiment is: RNA ≥ 300μg (plant tissue sample ≥ 2g), concentration > 350ng/µl, no EDTA, no DNA contamination. It is recommended to send 2~3 times the sample size for backup.

Project cycle

  • It takes 60~90 working days to complete the standard process.

Feedback to customers

  • Raw data
  • A project report includes experimental process, instrument parameters, analysis methods and results, etc.
