DAP seq in Plant

DAP-seq (DNA Affinity Purification sequencing) constructs and expresses transcription factor (TF) protein in vitro, binds to target genome fragments, and enriches the genomic DNA fragments bound to the target protein. Through the combination with high-throughput sequencing technology, the DNA products after DAP are sequenced and analyzed, and the DNA binding sites of the target protein are found from the whole genome, and high-throughput data results can be obtained by high-efficiency sequencing methods. DAP can effectively solve the limitation of CHIP technology's lack of specific antibodies for the target protein, and has a wider range of applications. The application of DAP-seq technology in the field of plant molecular breeding mainly focuses on studying the binding sites and functions of transcription factors, judging the type of histone modification at a specific position in the DNA chain, studying the relationship between histone covalent modification and gene expression, etc. It provides theoretical evidence for academic research projects that explain environmental stress, disease resistance, etc.
