Plant Genome Editing with TALEN
INQUIRYIntroductions
TALEN (transcription activator-like effector nuclease) is an artificially modified restriction endonuclease, composed of a DNA recognition domain and an endonuclease cleavage domain (Fok I). The principle of TALEN technology is not complicated. First, the DNA recognition element of TALEN binds to the target DNA site. Secondly, under the action of FokI nuclease, the specific site is cut. Finally, with the help of the inherent homology-directed repair (HDR) or non-homologous end joining pathway (NHEJ) in the cell, specific sequences are repaired, that is gene knockout, gene knock-in, and specific site mutation. Today, TALEN technology has been widely used in corn, rice, wheat, barley and other crops. The targeted modification of endogenous genes in crops by TALEN gene editing technology provides a new way for crop genetic improvement. Using this technology to delete large chromosomal segments can quickly knock out gene clusters that control undesirable traits, and achieve rapid genetic improvement of crops.
